I. Overview
FireGene Animal Tissue Storage Kit (Cat #. FG-BA3301) is designed to maintain the short-term viability of animal tissue cells. Under low-temperature conditions of 2°C to 8°C, this solution can preserve tissue cell activity for at least 72 hours, facilitating sample transportation and experimental scheduling. Simply immerse the sample completely in the preservation solution for easy operation. With clear components and stable performance, it effectively reduces sample loss before experiments and improves experimental success rates, making it an ideal choice for maintaining tissue cell viability.
The solution covers dozens of commonly used experimental species, including humans and mice, and is suitable for hundreds of tissue types, such as brain, liver, lung, stomach, intestine, kidney, tumor tissue, breast, ovary, fallopian tube, uterus, testis, epididymis, pituitary gland, thyroid gland, thymus, esophagus, nasal tissue, mucosa, periosteum, skin, lymphoid tissue, embryonic tissue, cartilage, etc.

III. Test Data
Experiments on fresh mouse brain and kidney tissues showed that the indicators of fresh tissues dissociated after 72 hours of preservation can still meet the requirements of single-cell transcriptome sequencing experiments.
III.1 Test Process
4 copies each of fresh mouse brain and kidney tissues were preserved with FireGene Animal Tissue Universal Preservation Solution for 24h, 48h, and 72h respectively, while one copy was unpreserved (0 h). Then they were dissociated using FireGene Mouse Brain Tissue Dissociation Kit and Kidney Tissue Dissociation Kit, and processed with aunSia automatic equipment.
III.2 Test Results
Quality inspection results of single-cell suspensions dissociated from mouse tissues preserved for different times are as follows:
For brain tissue:
At 0h, the viability cell rate is 98.72%, the clumping rate is 3.08%, and the nucleated rate is 94%.
After 24h of preservation, the viability cell rate is 94.05%, the clumping rate is 4.49%, and the nucleated rate is 91%.
After 48h, the viability cell rate is 90.77%, the clumping rate is 4.73%, and the nucleated rate is 90%.
After 72h, the viability cell rate is 87.81%, the clumping rate is 5.71%, and the nucleated rate is 90%.
For kidney tissue:
At 0h, the viability cell rate is 97.43%, the clumping rate is 3.07%, and the nucleated rate is 92%.
After 24h of preservation, the viability cell rate is 95.66%, the clumping rate is 2.96%, and the nucleated rate is 94%.
After 48h, the viability cell rate is 90.27%, the clumping rate is 4.15%, and the nucleated rate is 91%.
After 72h, the viability cell rate is 86.19%, the clumping rate is 4.36%, and the nucleated rate is 88%.
IV. Project Field Test
So far, we have completed a large number of single-cell sequencing projects on samples treated with this tissue preservation solution, involving dozens of tissue types!
IV.1 Data Performance of Partially Tested Tissues after Preservation
For human digestive system tissue preserved for approximately 48h, the cell viability is 90.22%, the clumping rate is 6.16%, the nucleated rate is 77%, the captured cell count is 14,639, and the median genes is 1,804.
For human tumor tissue preserved for approximately 48h, the cell viability is 89.21%, the clumping rate is 4.81%, the nucleated rate is 78%, the captured cell count is 9,766, and the median genes is 3,225.
For human respiratory system tissue preserved for approximately 72h, the cell viability is 92.06%, the clumping rate is 3.88%, the nucleated rate is 87%, the captured cell count is 15,558, and the median genes is 1,888.
For mouse nervous system tissue preserved for approximately 48h, the cell viability is 90.03%, the clumping rate is 3.91%, the nucleated rate is 90%, the captured cell count is 16,927, and the median genes is 2,444.
For mouse tumor tissue preserved for approximately 48h, the cell viability is 92.5%, the clumping rate is 5.37%, the nucleated rate is 81%, the captured cell count is 12,471, and the median genes is 2,320.
For mouse reproductive system tissue preserved for approximately 72h, the cell viability is 91.72%, the clumping rate is 2.94%, the nucleated rate is 85%, the captured cell count is 10,284, and the median genes is 1,950.
