Description
Product Description
This product utilizes a novel rapid transfer formulation, enabling the completion of Western Blot wet transfer in just 15–40 minutes. Anhydrous ethanol can be used instead of methanol during preparation; the process generates minimal heat, eliminating the need for an ice bath.
Product Features
Efficient and convenient:Wet transfer for Western blotting can be completed in 15–40 minutes; heat generation is minimal, eliminating the need for an ice bath;
Operational Safety:During preparation, anhydrous ethanol may be used instead of methanol to avoid the use of toxic reagents;
Good compatibility:Suitable for the transfer of various common gels.
Preparation Method(1×)
1000 mL Rapid Transfer Buffer (1×):
100 mL Rapid Transfer Buffer (10×) + 800 mL deionized water + 100 mL absolute ethanol (or methanol)
Note: During preparation, first dilute the Rapid Transfer Buffer (10×) with deionized water, and add the alcohol last.
Reference conditions for membrane transfer
Note: This product requires a constant current of 400 mA for the membrane transfer process.
1. Gradient gel (4%–15%, 1.5 mm thick), three-color prestained molecular weight marker WJ103 (10–250 kDa); transfer performed at a constant current of 400 mA for 30 minutes, resulting in the transfer of all bands.
2. For standard PAGE gels, transfer efficiency depends on gel concentration. Using a constant current of 400 mA, proteins smaller than 150 kDa can be completely transferred from 1.0 mm gels of various concentrations within the durations listed below; for proteins larger than 150 kDa, the transfer time should be extended by 5–10 minutes:
|
Gel concentration |
6% |
7.5% |
10% |
12.5% |
|
Recommended duration |
15 min |
20 min |
30 min |
35 min |
3. The greater the PAGE gel thickness, the lower the transfer efficiency; for 1.5 mm thick gels, the transfer time specified in Item 2 can be extended by 5–10 minutes.
Precautions
1. If the transfer duration is 45 minutes or less, do not use an ice bath, as this will reduce transfer efficiency; if the duration exceeds 45 minutes, the temperature may become excessively high, so using an ice pack to lower the temperature is recommended to ensure optimal experimental results;
2. For your safety and health, please wear a lab coat and disposable gloves while performing operations.
